Why is Lanadelumab-flyo Prescribed? (Indications)

Lanadelumab-flyo injection is prescribed to treat type I and type II hereditary angioedema (HAE) in patients aged 12 years and above.

Hereditary angioedema leads to repeated episodes of severe swelling of limbs, face, intestinal tract, and airway and can be fatal. It is a severe and rare genetic disease found in patients with either low levels or improper functioning of a specific protein (C1-INH) in the body.

HAE affects about 1 in 10,000 to 1 in 50,000 people. Type I HAE is the most common comprising of 85 % of the reported cases. Some patients have many attacks in a single month while some patients can survive free of attacks for months.

Lanadelumab-flyo is a human monoclonal antibody that works by blocking a specific enzyme called plasma kallikrein, thereby preventing the swelling and pain attacks of hereditary angioedema from re-occurring.

When should Lanadelumab-flyo not be taken? (Contraindications)

Lanadelumab-flyo should not be used in patients with-

• If there is an allergy to lanadelumab-flyo
• Children below 12 years of age

What is the dosage of Lanadelumab-flyo?

• The initial recommended dose of lanadelumab-flyo is 300 mg given every two weeks.

• The dosing interval of lanadelumab-flyo can also be given as 300 mg for every four weeks if the patients have not reported any attacks or the symptoms of hereditary angioedema are under control.

How should Lanadelumab-flyo be taken?

• Lanadelumab-flyo is available as an injection form which should be given just under the skin or subcutaneously.

• The injection can be self-administered or taken with the help of a caregiver. The caregiver or the patient should take adequate training from a healthcare provider before administering an injection.

• Lanadelumab-flyo injection comes in a single-dose vial as a ready-to-use solution that does not require any additional dilution or reconstitution before administration.

• The injection vial should be removed from the refrigerator and the temperature of the vial should be allowed to rise to the room temperature. This will take about 15 minutes.

• The injection sites are the upper arm, abdomen, or thigh and the injection should be administered within 10 to 60 seconds.

• After withdrawing the prescribed dose of lanadelumab-flyo by a dosing syringe, it should be injected within 2 hours or can be used within 8 hours if stored in a refrigerator between 2°C to 8°C.

• Discard if any unused portions of lanadelumab-flyo are left in the vial or syringe after the scheduled storage time has passed.

What are the warnings and precautions for Lanadelumab-flyo?

• Severe allergic reactions can occur during the course of lanadelumab-flyo treatment and patients must be advised to consult the physician if such allergic or hypersensitivity reactions occur.

• During severe allergic episodes, treatment with lanadelumab-flyo should be discontinued and the patient must be treated appropriately with proper medical care.

• Caution is required while treating pregnant or breastfeeding mothers with the lanadelumab-flyo injection to minimize the harm caused to the fetus or the baby.

• Do not inject without proper training from the healthcare personnel and never give the medicine to another person who has similar symptoms of hereditary angioedema as it may harm them.

• The injection sites can be changed frequently to prevent the formation of hardened tissue or lumps.

What are the side effects of Lanadelumab-flyo?

The most common side effects of lenadelumab-flyo injection are-

• Injection site reactions (Pain, swelling, itching, bleeding, bruising)
• Headache
Dizziness or lightheadedness
• Upper respiratory infections (Cough, nasal congestion, sore throat)
• Loose stools
Rashes
• Muscle pain
• Abnormal levels of liver enzymes

What are the other precautions for Lanadelumab-flyo?

• Check the lanadelumab-flyo vial for the presence of any discoloration or visible particles before injecting. Discard immediately if such things are found.

• Do not inject lenadelumab-flyo into hardened, inflamed, injured, or bruised skin.

• Also, avoid shaking the vial vigorously.
