Larotrectinib is an anticancer drug(kinase inhibitor) that is used to treat solid tumors in adults and children having certain gene modifications. Solid tumors are an abnormal mass of tissue that does not contain liquid areas and cysts. 

Why is Larotrectinib Prescribed? (Indications)

This medication is an anticancer agent that is indicated for the treatment of solid tumors in adults and children. It is specifically used in patients having gene modifications like NTRK gene fusion.

When should Larotrectinib not be taken? (Contraindications)

1.Allergy to the drug
2.Pregnant women
3.Breastfeeding women

What is the dosage of Larotrectinib?

For adults and children- Capsule- 25mg, 100mg.  Oral solution- 20mg/mL. The dose recommended of larotrectinib is 100mg orally twice daily with or without food for atleast 6 months.

How should Larotrectinib be taken?

Capsules and solutions are available which can be taken by orally. For oral solution, always use the syringe given to measure the dose.

Missed a Dose?

Do not take a missed dose if the next dose is to be taken within 6 hours. If the person vomits after administration of a dose, next dose should be taken according to the schedule.

What are the side effects of Larotrectinib?

1.General: Fatigue, swelling of lower legs or hands, fever
2.Gastrointestinal: Nausea, vomiting, constipation, diarrhea, abdominal pain
3.Nervous system: Dizziness, headache
4.Respiratory: Cough, nasal congestion, shortness of breath
5. Vascular: Hypertension
6.Muscles and Joints: Back pain, joint pain, muscle pain, pain in the extremities, muscle weakness.

What are the other precautions for Larotrectinib?

1.Monitor liver function tests every two weeks during the first month and after that monthly. Reduce the starting dose of larotrectinib in patients with moderate liver function impairment.
2.Neurological adverse effects like gait disturbances, dizziness, tremors, delirium, memory impairment can be seen.
3.Patients are advised not to operate heavy machines if they experience these adverse effects.
4. In females who are able to get pregnant, proper contraceptives should be used during the treatment and one week after the last dose is given.
5. In males with female partners who are able to pregnant, proper contraceptives should be used during the treatment and one week after the last dose is given.
6.Patients should inform their doctors about all the medications they take including herbal medications, OTC products, and vitamin supplements.
7.Do not eat grapefruit or drink grapefruit juice.
Do not take St John Wort.
