Why is Olaratumab Prescribed? (Indications)

Olaratumab belongs to a group of medications called monoclonal antibodies. It is a platelet-derived growth factor receptor alpha (PDGFR-α) blocking antibody used along with doxorubicin to treat certain types of soft tissue sarcomas in adults that cannot be treated with surgery or radiation. Soft tissue sarcomas are cancers that originate from tissues like muscle, fat, tendons, blood vessels or nerves.

The drug mainly acts by slowing or stopping down the growth of cancer cells.

When should Olaratumab not be taken? (Contraindications)

Currently, there are no definite contraindications to the use of olaratumab drug.

What is the dosage of Olaratumab?

Olaratumab is administered in a dosage of 15 mg/kg as a slow intravenous injection on day 1 and day 8 of the 21-day chemotherapy cycle. It is administered along with the anticancer drug doxorubicin for the first 8 cycles. The course of the treatment depends on how well the patient responds to the treatment.

How should Olaratumab be taken?

Olaratumab is administered as an intravenous infusion diluted in 0.9% sodium chloride.The injection is administered slowly over an hour.

What are the warnings and precautions for Olaratumab?

• Avoid getting pregnant during olaratumab treatment as well as until 3 months after your final dose as it can harm the baby.

• Consult your doctor immediately if you are allergic to the injection or to any other medications while taking olaratumab. Premedication with the anti-histamine diphenydramine and the corticosteroid dexamethasone is required before administering olaratumab for the first time to prevent an allergic reaction.

• Inform your doctor if you experience any infusion-related reactions, which will require immediate stopping of the medication.

• Abnormal laboratory test results have been noted in patients being administered olaratumab like reduced platelet and white blood cell counts, high blood glucose levels and low potassium and phosphate levels.

Gastrointestinal - Nausea, vomiting, diarrhea, abdominal pain, loss of appetite, mouth sores.
Skin - Hair loss
Central nervous system - Anxiety, headache, neuropathy (problem with the nerves)
