Macimorelin comes as granules in the 60mg sachet which has to be reconstituted with 120 ml of water and should be taken by mouth.
• Patients weighing below 120 kg - Use 1 sachet
• Patients weighing above 120 kg - Use 2 sachet
• The reconstituted solution should be used within 30 minutes after preparation and the unused solution should be discarded.
Test procedure:
• The recommended dose of macimorelin granules can be determined by multiplying the patient weight in kilogram (0.5mg/kg).
• Dissolve the contents of the sachet appropriately in the required volume of water and stir the contents gently for 2 to 3 minutes to obtain a final concentration of 0.5mg/ml.
• An appropriate volume of the solution can be transferred by using a syringe without the needle into a drinking glass which should be drunk in 30 seconds.
• The blood samples should be withdrawn at regular time intervals of 30, 45, 60 and 90 minutes after the administration of macimorelin oral solution to determine the concentration of growth hormone.
• If the serum growth hormone level is less than 2.8 ng/ml at all the measured time points after macimorelin administration, the test result confirms the presence of adult growth hormone deficiency.
• It is taken as a single dose of 0.5mg/kg body weight after a fasting period of 8 hours.

• Deficiencies of sex hormones, thyroid hormones and/or glucocorticoids
• Patients taking drugs that can affect pituitary GH secretion
